Fusarium wilt (FW) disease of cotton, caused by the fungus oxysporum f. sp. vasinfectum (Fov), causes severe losses in cotton production worldwide. Though significant advancements have been made development FW-resistant Upland (Gossypium hirsutum) resistance screening programs, precise genes and corresponding molecular mechanisms for to Fov remain unclear. Herein it is reported that Fov7, a gene unlike canonical plant disease-resistance (R) genes, putatively encoding GLUTAMATE RECEPTOR-LIKE (GLR) protein, confers race 7 cotton. A single nucleotide polymorphism (SNP) (C/A) GhGLR4.8, resulting an amino acid change (L/I), associated with resistance. PCR-based DNA marker (GhGLR4.8SNP(A/C)) developed shown cosegregate CRISPR/Cas9-mediated knockout Fov7 results lines extremely susceptible loss ability induce calcium influx response total secreted proteins (SEPs) Fov.
